چکیده

Natural selection is believed to be an unforgiving and relentless force in the evolution of life on earth. An organism that cannot adapt to a changing environment or an environment hostile to cell functions is at risk as a species. So it is important to understand the mechanisms used by plants, animals, and microorganisms in adapting to environments in the biosphere that would ordinarily denature proteins or otherwise cause disruption of life-giving cellular processes. These hostile environments involve such stresses as extremes of temperature, cellular dehydration, desiccation, high extracellular salt environments, and even the presence of denaturing concentrations of urea inside cells (1). It has been recognized for some time that many plants, animals, and microorganisms that have adapted to environmental extremes also accumulate significant intracellular concentrations of small organic molecules (1–4). From these (and other) observations comes the hypothesis that these small organic molecules, called osmolytes, have the ability to protect the cellular components against denaturing environmental stresses (1–5). In this chapter, we seek to understand the molecular-

Biological Wonders of Osmolytes: The Need to Know More

Nature has selected osmolytes to protect intracellular macromolecules against denaturing stress conditions. These molecules are accumulated in the intracellular environment at considerably high concentrations. In general, osmolytes are known to stabilize proteins. However, under certain conditions, their destabilizing properties have also been pointed out. A careful qualitative and quantitative...

Osmolyte solutions and protein folding

In this brief review we discuss the evolution of recent thought regarding the role and mechanism of osmolytes with respect to protein stability. Osmolytes are naturally occurring intracellular compounds that change the protein folding landscape. Contributions from experiments are considered in the context of current theory and simulation results.

Osmolyte-Induced Folding and Stability of Proteins: Concepts and Characterization

It is well-known that the typical protein’s three-dimensional structure is relatively unstable in harsh conditions. A practical approach to maintain the folded state and thus improve the stability and activity of proteins in unusual circumstances is to directly apply stabilizing substances such as osmolytes to the protein-containing solutions. Osmolytes as natural occurring organic molecules ty...

Protein thermal stabilization in aqueous solutions of osmolytes.

Proteins' thermal stabilization is a significant problem in various biomedical, biotechnological, and technological applications. We investigated thermal stability of hen egg white lysozyme in aqueous solutions of the following stabilizing osmolytes: Glycine (GLY), N-methylglycine (NMG), N,N-dimethylglycine (DMG), N,N,N-trimethylglycine (TMG), and trimethyl-N-oxide (TMAO). Results of CD-UV spec...
